摘要:
目的经皮犬传染性性病肿瘤(CTVT)细胞悬液注射法建立犬同种移植性肺癌模型并研究其生物学特性。方法纯种比格犬12只,接种前4d口饲环孢素。腹腔麻醉后,CT引导下穿刺至犬肺中叶或后叶,注入1.0mlCTVT细胞悬液(细胞活度:108/ml)。接种后即刻及第2、4、5、6、8、10周行胸部CT平扫,观察肿瘤生长及转移情况;每日观察动物的一般情况,包括精神状态、饮食、呼吸及活动等情况。待其自然死亡或于第10周处死,进行病理学检查。结果12只犬共接种15点。第5周CT扫描可见2只犬肺内成瘤,第6周9只犬肺内成瘤,观察至第10周共9只犬10个接种点肺内成瘤,接种点成瘤率66.7%。第6、8、10周肿瘤最大层面均径分别为(1.059±0.113)cm、(1.827±0.084)cm和(2.389±0.153)cm,最大直径达3.5cm;所有成瘤犬最后均出现中至重度胸腔积液及纵隔淋巴结肿大。结论经皮CTVT细胞悬液注射法可建立大动物肺癌模型,有助于肺癌的相关研究。
